The Antioxidant Effect of Curcumin on Miniature Pig Sperm Cryopreservation

Abstract
Cryopreservation is accompanied by rapid temperature changes. Boar spermatozoa are containing the high content of unsaturated fatty acids. For that reason, boar sperm sensitive to oxidative stress causes during frozen-thawing compare with another species. Thus antioxidant is essential substance of freezing buffer, we utilize curcumin for antioxidant of freezing buffer. Curcumin have been proven to be effective, is widely known now. The purpose of this study to determine the effects of curcumin added to freezing buffer on semen parameters, Lipid peroxidation (LPO) which is caused by reactive oxygen species (ROS) in sperm membrane and total sperm oxidation stress after post-thawing of boar sperm. Semen parameter is measured using CASA and determine ROS rate, oxidative stress of boar sperm using FACS. Boar semen samples diluted with freezing buffer containing curcumin (0, 10, 50, 100 and 500 ¥ìM) was cooled to 4 o C and frozen in 0.5 mL straws that post-thawing were evaluated. As a result, we found that curcumin could protect the sperm plasma membrane from free radical and LPO during boar sperm postthawing. While no additive (control) sample was showed motility 49.8¡¾4% and progressive motility 33.4¡¾2%, curcumin (10 ¥ìM) sample showed significant increases in motility 50.4¡¾2.9%, progressive motility 31.7¡¾4% (P<0.05). These results indicate that supplementing with curcumin (10 ¥ìM) during post-thawing sperm capacity preserve from oxidative stress and induce LPO of sperm membrane.
